Megatherium ( ; from Greek () 'great' + () 'beast') is an extinct genus of large ground sloths endemic to South America that lived from the Early Pliocene through the end of the Late Pleistocene. It is best known for the elephant-sized, type species Megatherium americanum, primarily known from the Pampas, but ranging southwards to northernmost Patagonia and northwards to southern Bolivia during the late Middle Pleistocene and Late Pleistocene. Glyptodon (; ) is a genus of glyptodont, an extinct group of large, herbivorous armadillos that lived from the Pliocene, around 3.2 million years ago, to the early Holocene, around 11,000 years ago, in South America. It is one of, if not the, best-known genera of glyptodont. Glyptodon has a long and storied past, being the first named extinct cingulate and the type genus of the subfamily Glyptodontinae. Thalassocnus is an extinct genus of semiaquatic ground sloths from the Miocene and Pliocene of the Pacific South American coast. It is monotypic within the subfamily Thalassocninae. The five species—T. antiquus, T. natans, T. littoralis, T. carolomartini, and T. yuacensis—represent a chronospecies, a population gradually adapting to marine life in one direct lineage. Acrophyseter is a genus of extinct sperm whale that lived in the Late Miocene off the coast of what is now Peru. The genus comprises two species: A. deinodon and A. robustus. It is part of a group of macroraptorial sperm whales that all share several features for hunting large prey, such as deeply rooted and thick teeth. Acrophyseter measured in length, making it the smallest macroraptorial sperm whale currently known. Because of its short pointed snout and strongly curved front teeth, it probably fed on the marine vertebrates of its time, such as seals and other whales.
Gryposuchus
Gryposuchus (meaning "hooked crocodile") is an extinct genus of gavialid crocodilian. Fossils have been found from Argentina, Colombia, Venezuela, Brazil and the Peruvian Amazon. The genus existed during the Miocene epoch (Colhuehuapian to Huayquerian). One recently described species, G. croizati, grew to an estimated length of . Gryposuchus is the type genus of the subfamily Gryposuchinae, although a 2018 study indicates that Gryposuchinae and Gryposuchus might be paraphyletic and rather an evolutionary grade towards the gharial.
Inkayacu
Inkayacu is a genus of extinct penguins. It lived in what is now Peru during the Late Eocene, around 36 million years ago. The only species, I. paracasensis, was named from a single nearly complete skeleton discovered in 2008. It includes fossilized feathers, the first known in penguins. A study of the melanosomes, pigment-containing organelles within the feathers, indicated that they were gray or reddish brown. This differs from modern penguins, which get their dark black-brown feathers from unique melanosomes that are large and ellipsoidal.

Peregocetus
Peregocetus is a genus of early whale that lived in what is now Peru during the Middle Eocene epoch. Its fossil was uncovered in 2011 in the Yumaque Member (Paracas Formation) of the Pisco Basin at Playa Media Luna by a team consisting of members from Belgium, Peru, France, Italy, and the Netherlands. Parts recovered include the jaw, front and hind legs, bits of spine, and tail. Olivier Lambert, a scientist at the Royal Belgian Institute of Natural Sciences and lead author of the study, noted that Peregocetus "fills in a crucial [knowledge] gap" about the evolution of whales and their spread.

Metaxytherium is an extinct genus of dugong that lived from the Oligocene until the end of the Pliocene. Fossil remains have been found in Africa, Europe, North America and South America. Generally marine seagrass specialists, they inhabited the warm and shallow waters of the Paratethys, Mediterranean, Caribbean Sea and Pacific coastline. American species of Metaxytherium are considered to be ancestral to the North Pacific family Hydrodamalinae, which includes the giant Steller's sea cow.

It lived in Argentina, Peru, and Venezuela between the middle Eocene and the Late Miocene, ~42–11.6 Ma. Described in 2007, based on a severely damaged specimen from which only a snout tip was recovered, Barinasuchus is known from a single species, B. arveloi, named after Alberto Arvelo Torrealba, a local educator and poet.

Notiomastodon
Notiomastodon is an extinct genus of gomphothere proboscidean (related to modern elephants), endemic to South America from the Pleistocene to the early Holocene. Individuals of Notiomastodon reached a size similar to that of the modern Asian elephant, with a body mass of 3-4 tonnes. Like other brevirostrine gomphotheres such as Cuvieronius and Stegomastodon, Notiomastodon had a shortened lower jaw and lacked lower tusks, unlike more primitive gomphotheres like Gomphotherium.
Mystacodon
Mystacodon is a genus of toothed baleen whale from the Late Eocene Yumaque Member of Paracas Formation (previously called as Yumaque Formation) of the Pisco Basin in southwestern Peru. It is the oldest known baleen whale, and was probably a suction feeder of small prey on the seafloor.
Parapropalaehoplophorus septentrionalis
Parapropalaehoplophorus is an extinct genus of comparatively small (compared to Glyptodon) species of glyptodont, extinct relatives of the modern armadillo. The sole species, P. septentrionalis, identified in 2007 from the fossilized remains of a specimen found in 2004, weighed approximately 200 pounds and had a shell covered by tiny circular bumps. It lumbered around northern Chile in the Chucal Formation, an area now dominated by the Andes mountain range, some 18 million years ago. Fossils of the glyptodont also have been found in Peru (Ipururo and Pebas Formations).

Supayacetus
Supayacetus is an extinct genus of basilosaurid cetacean from the Middle Eocene (Bartonian stage) Paracas Formation of Peru. It has been noted for its relatively small size and basal morphology, with the sternum bearing close resemblance to those of protocetids. Due to this, it has been traditionally placed as one of the basalmost basilosaurids, except for a 2023 study that places it within the family Pachycetinae close to Neoceti. Supayacetus is monotypic, meaning the genus includes only a single species: S. muizoni.

Rhynchippus ("beaked horse") is an extinct genus of notoungulate mammals from the Late Oligocene (Deseadan in the SALMA classification) of South America. The genus was first described by Florentino Ameghino in 1897 and the type species is R. equinus, with lectotype MACN A 52–31. Fossils of Rhynchippus have been found in the Agua de la Piedra and Sarmiento Formations of Argentina, the Salla and Petaca Formations of Bolivia, the Tremembé Formation of Brazil, and the Moquegua Formation of Peru.

Berriasella is a discoidal evolute perisphinctacean ammonite, and type genus for the neocomitid subfamily Berriasellinae. Its ribbing is distinct, consisting of both simple and bifurcated ribs that extend from the umbilical seam across the venter; its whorl section generally compressed, the venter more or less narrowly rounded. Gnatusuchus is an extinct genus of caiman represented by the type species Gnatusuchus pebasensis from the Middle Miocene Pebas Formation of Peru. Gnatusuchus lived about 13 million years ago (Ma) in a large wetland system called the Pebas mega-wetlands that covered over one million square kilometers of what is now the Amazon Basin (the modern basin had not yet developed at that time and instead of draining from west to east into the Atlantic Ocean, river systems drained northward through the wetlands and into the Caribbean Sea).

Miocochilius is an extinct genus of small notoungulate mammals (typotheres) native to South America. The genus lived during the Middle Miocene epoch (Laventan in the SALMA classification). The genus contains two described species, the type species M. anomopodus described in 1953 by Ruben Arthur Stirton and M. federicoi, described and included in the genus by Darin A. Croft.

Piscobalaena is an extinct genus of cetaceans, which lived from the Middle to Late Miocene epochs (about 11.6 to 5.3 million years ago) in Peru and Florida. The type species, H. martini, was found in the Pisco Formation, together with other marine animals such as crustaceans, sharks, coastal birds, whales and aquatic sloths. The distinguishing feature of the seal was its teeth: they were extremely robust, hence the name (, "stout" in Greek; , tooth in Quechua).
